Abstract:

The influence of BAP (0.1; 0.5 mg 1') and meto-topolin (mT) (0.2; 1.0 mg I ’) added alone or together with IBA (0.02 mg M)i on the growth and development of shoot tips and axillary buds of P. x hortorum ‘Bargpalais’ isolated in April and July was compared. The explants taken in April had higher regeneration ability. Both cytokinin stimulated growth of buds, but shoots of good quality (without deformation and vitrification symptoms) were noted on the medium supplemented with mT (0.4 mg I ')- Application of mT (1.0 mg I ') in the multiplication stage resulted in better shoots quality as well as more effective shoots formation (4.8-7.1, depending on subculture) compared to BAP. The observation of 4 subsequent subcultures after initiation of cultures showed that the quality of shoots P. X hortorum grown in the presence of BAP decreased with time.
