Abstract:

Thrombin inhibitors are anticoagulants that inhibit clot formation. The most potent, knownthrombin inhibitor is hirudin, a 65 amino acid protein isolated from the salivary glands of themedicinal leech. An X-ray three — dimensional structure of the complex of hirudin with humana-thrombin has been recently reported. This review article describes the design based on thecrystal structure, of new synthetic hirudin analogs with reduced sequence and enhanced proteolytic stability.
