Abstract:

Activity of alkaline phosphatase and adenylate cyclase was evaluated in the brain vascularnetwork of rats subjected to 10 min clinical death due to cardiac arrest.Progressive decrease of both enzymes activities was noted in the early survival stages, reachingthe lowest intensity in 24th h after ischemic incident. Restitution of enzymatic activity was observed2 days later, although even then it did not reach intensity typical for normal controls. Electronmicroscopic study revealed disturbances of normal polarity of enzyme distribution in thecytoplasmic membranes of vascular endothelia. Activity of enzymes disappeared from luminalendothelial surfaces, appearing on abluminal ones and in the capillary basal membranes.Ultrastructural picture of the cerebral capillaris was additionally characterized by remarkableincrease of micropinocytic vesicles, and appearance of coated vesicles as well as canalicularstructures. Swelling of endothelial cytoplasm and widening of interendothelial clefts were alsopresent.The results obtained indicate that deep 10 min ischemia in experimentally induced clinicaldeath is leading to the impairment of transport functions in damaged cerebral capillaryvessels.
