The methods of evaluation of thermodynamic parameters for oligonucleotide order-disordertransitions are presented. These parameters are useful for the prediction of stability of duplexesformed by these oligonucleotides with RNA matrix. The common opinion, that better antisenseproperties can be achieved by the increase of oligonucleotide length is critically evaluated. Theproblem of optimal length of oligonucleotide for antisense purposes is discussed by taking intoaccount the selectivity of hybridization.
