Abstrakt:

Results of morphologic observations on cultures of glial tissue from neonate rats and mice and chick embryos are reported. The cultures were grown in Carrel flasks and on the double cover slips by the method of Maximow. Observations were made on fixed cultures, which had survived for 1 —58 days. Three phases in the growth of the cultures were distinguished, differing with respect to type of prevailing cells, their maturity, mitotic indexes, and distribution of the cel population. The phases were as follows: I—growth of undifferentiated cells (1—4days), II—differentiation of the cells (uptothe14thday),III—growth of the differentiated culture (after the second week). The morphologic criteria of differentiation of glial cells in fixed stained preparations are discussed. The need of this differentiation arises from the use of cultures in histochemical studies. The features differentiating astroglia from oligodendroglia include dimensions, shape and stainability of the nucleus, character of the protoplasm and manner of formation of processes. The morphology of different types of glial cells observed in culturesis discussed, including the various types of giant cells which were encountered. The basic type of cel division observed in cultures was indirect division, which occurs in very early stages of the growth of cultures. This type of division, beside emigration of cells from the explantate, plays the main role in the multiplication of the cel population in the early stages of growth. The tempo of development of the culture expressed in terms of mitotic indexes varies in different stages of development of the culture, being highest during the stage of differentiation of the cells, and lowest during growth of the differentiated culture. The marked morphologic variation of cells of the same type, presumably due to different functional phases, is emphasized. As a result of this variation, part of the cells in fixed stained preparations cannot be classified.
